10x Visium Spatial

10x Visium Spatial is a platform for profiling RNA expression in tissue, with the ability to profile at single-cell and sub-cellular resolution while maintaining spatial context. The 10x Visium Spatial is a spatial transcriptomics platform for profiling FFPE, fresh frozen, and fixed frozen tissues, without destroying the tissue sample.

Some of the available assays include Spatial Gene Expression v1 & v2, which contain the whole transcriptome (various species); Spatial Gene & Protein Expression, which contains the whole transcriptome with the ability to co-detect 31 proteins (human or mouse); and HD Spatial Gene Expression, which enables profiling at single-cell and sub-cellular resolution and contains the whole transcriptome (human or mouse).

profiling using arrays of spatially-barcoded probes

The Visium Spatial platform uses assay-specific slides containing arrays of spatially barcoded spots, each with millions of probes targeting the whole transcriptome.

The basic workflow involves mounting tissue over an array, adding the whole transcriptome probes to the tissue. The tissue is stained and imaged, followed by hybridization of the probes to target genes in the tissue, which are then ligated. The resulting ligation products are released from the tissue and bind locally to the spatially barcoded spots on the slide. cDNA libraries are then created from the spatially barcoded probes and subsequently sequenced.

sample requirements

  • Compatible samples: FFPE, Fresh Frozen, or Fixed Frozen tissue.
  • Recommended section thickness:
    • FFPE, FF, or FxF: 10 µm is used for most tissue types
