GeoMx® Digital Spatial Profiler
The GeoMx DSP is a platform for profiling RNA and protein expression in distinct cell populations or compartments while maintaining spatial context. The GeoMx DSP is a spatial multiomics platform for expression profiling FFPE, fresh frozen, and fixed frozen tissues, without destroying the tissue sample.
Some of the available assays include the GeoMx Whole Transcriptome Atlas containing over 20,000 genes (human or mouse), the GeoMx Cancer Transcriptome Atlas containing over 1,800 genes (human or canine), and the GeoMx IO Proteome Atlas containing over 570 protein targets.
For a full overview of available assays see:

Flexible Profiling by Combining IHC and ISH
The GeoMx DSP instrument is equipped with four fluorescent imaging channels: FITC, Cy3, TexasRed, and Cy5. Using immunohistochemical techniques, fluorescently labeled antibodies and/or RNA probes are used to stain FFPE, fresh frozen, or fixed frozen tissues.
Through in situ hybridization, GeoMx probes are hybridized to targets in the tissue. Once tissue structures are visualized by the fluorescently labeled antibodies, distinct cell populations or compartments can be selected by the user as regions of interest (ROI). The maximum size for each ROI is 660 x 785 µm.
Using the GeoMx DSP software, ROIs are then segmented into biological compartments called areas of illumination (AOI), from which the GeoMx probes are collected for digital profiling.
Sample requirements
- Compatible samples: FFPE, Fresh or Fixed Frozen tissue.
- Tissue should be sectioned within 4 weeks of planned GeoMx experiment.
- We recommend mounting tissue sections on TOMO slides.
- Recommended section thickness
- FFPE: 5 µm
- FF or FxF: 5-10 µm
- Tissue should be mounted within the GeoMx scannable area of 14,1 x 35,3 mm, shown
with green in the figure below. - After sectioning the slide should dry overnight at room temperature.
- Once dried, the slide should be stored at 4C with desiccant.
